摘要
目的探讨MTS1与胃癌的关系。方法32例胃癌和30例正常胃粘膜标本提取细胞DNA、总RNA,MTS1cDNA探针采用随机引物法进行32PdATP掺入标记,MTS1与所提DNA、RNA进行Southern和Northern杂交分析,检测标本中有无MTS1的缺失和mRNA的不表达。结果MTS1Southern和Northern杂交分析结果显示,32例胃癌标本MTS1基因缺失及mRNA不表达率分别为34.4%和40.6%,明显高于正常胃粘膜组。MTS1基因缺失及mRNA不表达与胃癌浸润的深度及分化程度有关,胃癌侵及肌层组MTS1基因缺失及mRNA不表达率分别为52.6%和57.9%,胃癌未侵及肌层组分别为7.7%和15.4%,高、中分化胃癌组MTS1基因缺失及mRNA不表达率分别为7.1%和14.3%,而低分化及印戒细胞胃癌组分别为55.8%和61.1%。结论胃癌的发生、发展与MTS1基因的缺失及mRNA不表达有关,检测胃癌组织中MTS1变化对胃癌的诊断,了解胃癌的生物学特征,为病人预后的评估及治疗提供新的有价值的客观依据。
Objective To appraise the retationship between MTS1 gene and gastric carcinomas. Methods The cellular DNA and total RNA were isolated from samples of 32 gastric cancer and 30 normal gastric mucosa. MTS1 cDNA probes were labeled randomly by priming with 32 P dATP. MTS1. Southern and Northern blots were used to detect whether MTS1 gene deletion and MTS1 mRNA were unexpressed in the samples.Results MTS1 Southern and Northern blots showed that the deletion rate of MTS1 gene and unexpression rate of MTS1 mRNA were 34.4% and 40.6% in 32 samples of gastric cancers respectively, evidently higher than those of normal gastric mucosa group. Loss of MTS1 gene and unexpression of MTS1 mRNA were related to the depth of invasion and grade of differentiation of gastric cancer. The deletion rate of MTS1 gene and unexpression rate of MTS1 mRNA were 52.6% and 57.9% in gastric cancer invading muscular layer, 7.7% and 15.4% in non invading group, respectively. The deletion rate of MTS1 gene and unexpression rate of MTS1 mRNA were 7.1% and 14.3% in highly and moderately differentiated gastric cancer, 55.6% and 61.1% in poorly differentiated and signet ring cell carcinomas, respectively.Conclusion The occurrence and development of gastric cancer were related to the deletion rate of MTS1 gene, unexpression of MTS1 mRNA. Detecting alteration of MTS1 gene in tissue of gastric cancer provides rational basis for diagnosing gastric cancer, understanding of the biological characteristic, assessing the prognosis and treatment.
