摘要
目的:探讨仙鹤草丙酮提取物正丁醇萃取层对肝癌BEL-7402细胞的增殖抑制作用及其作用机制。方法:以不同浓度的仙鹤草丙酮提取物正丁醇萃取层作用于体外培养的BEL-7402细胞,MTT法检测细胞生长抑制率,应用流式细胞仪检测细胞凋亡率。应用Fluo-3/AM荧光探针观察细胞内钙离子浓度的变化,并用流式细胞仪检测细胞内活性氧的变化。结果:仙鹤草丙酮提取物正丁醇萃取层可显著抑制BEL-7402细胞的生长,诱导细胞发生凋亡,其IC50为107.54μg/mL。仙鹤草丙酮提取物正丁醇萃取层在作用48h后BEL-7402细胞数量明显的减少,在Fluo-3/AM荧光探针的作用下有较强的绿色荧光,同时检测出细胞内ROS有较明显的增加。结论:仙鹤草丙酮提取物正丁醇萃取层能抑制BEL-7402细胞的生长,诱导细胞发生凋亡;细胞内钙离子的释放和细胞内ROS的增加可能是其作用机制。
Objective:To investigate the antiproliferation effects of N-butanol layer of acetone extract of agrimony on hepatocellular carcinoma BEL-7402 cells and its mechanisms of action.Methods:BEL-7402 cells in culture medium were treated with different concentrations of N-butanol layer of acetone extract of agrimony.The inhibitory rate of the cells was measured by MTT assay.Cell apoptotic rate was detected by flow cytometry(FCM).Fluo-3 /AM fluorescent probe was used to observe the Ca2 + variation in the tumour cells.The variation of intracellular ROS was detected by FCM.Results:The N-butanol layer of acetone extract of agrimony could inhibit the growth of BEL-7402 cells and cause apoptosis significantly.The IC50 was 107.54 μg/mL.The number of cells was decreased especially after the cells were treated 48 h by the N-butanol layer of acetone extract of agrimony,and the green fluorescence was saw clearly by Fluo-3 /AM fluorescent probe.The intracellular ROS was increased obviously.Conclusions:The N-butanol layer of acetone extract of agrimony could inhibit the growth of BEL-7402 cells and cause apoptosis.The release of the intracellular Ca2 + variation and the increasing of the intracellular ROS may be its anti-hepatocellular carcinoma mechanisms.
