中脑多巴胺受体亚型在针刺减轻海洛因欣快记忆激发中的作用 被引量:2

Effect of Subtypes of DA Receptors in Mesocerebrum Acting in Acupuncture Obstructing Provocation of Euphoria Memory

摘要 目的:研究针刺减轻欣快记忆激发的中脑多巴胺受体机制。方法:将无天然位置偏爱的90只雄性昆明种小鼠随机分为空白组、针刺空白组、模型组、模型针刺腧穴组、模型针刺非穴组、模型纳曲酮组,造海洛因康复期心理依赖模型,选用低频电针作用于实验小鼠的"三阴交"、"内关"、"四神聪",每日1次,10次为1疗程;采用免疫组化检测中脑DA受体亚型D1、D2、D3、D4活性,用原位杂交法检测D1、D2、D3、D4mRNA表达的变化。结果:小鼠造模后,中脑DA受体亚型D1、D2、D3、D4活性极显著性升高(P<0.01),受体相应的mRNA表达极显著性增多(P<0.01);治疗后针刺腧穴组的D1、D2、D3活性极其mRNA表达均降低(P<0.01),针刺腧穴组D1、D2、D3活性及其mRNA表达显著低于针刺非穴组(P<0.05),针刺腧穴组D1、D2活性极其mRNA表达显著低于纳曲酮组(P<0.01)。治疗后模型组、模型针刺腧穴组、模型针刺非穴组、模型纳曲酮组间D4活性极其mRNA表达水平无显著性差异(P>0.05)。结论:针刺减轻海洛因心理依赖小鼠欣快记忆的激发,主要通过下调中脑多巴胺受体亚型D1、D2、D3的活性及其mRNA表达来实现。针刺腧穴组与针刺非穴组相比对D1、D2、D3活性极其mR-NA表达下调作用更显著,针刺腧穴组与纳曲酮相比对D1、D2活性极其mRNA表达下调作用更显著。D4可能没有参与针刺减轻欣快记忆激发途径。 Objective:To study the mechanism of Dopamine receptors in mesocerebrum on acupuncture acting obstructing the provocation of heroin euphoria memory.Methods:90 male mice were be randomly divided into 6 groups afterfiltration by CPP,the groups were: blank control group(A),blank with acupoints control group(B),model group(C),model with acupoints group(D),model with non-acupoint group(E),naltrexone group(F).The model mice of Heroin psychological dependence were formed by giving heroin with increasing dosage day by day.Mice were treated by acupuncture on acupoints as "Neiguan","Sanyinjiao","Sishengchong" once a day and lasted for 10 days.The activity of D1、D2、D3、D4 were measured by immumohistochemistry(IH) and the expression of D1 mRNA、D2 mRNA、D3 mRNA、D4 mRNA were detected by in situ hybridization histochemistry.Results:The activity of D1、D2、D3、D4 and the expression of the D1 mRNA、D2 mRNA、D3 mRNA、D4 mRNA increased in model mice with statistical significance(P0.01).The activity of D1、D2、D3 and the expression of the D1 mRNA、D2 mRNA、D3 mRNA of the model with acupoints group were lower than the model with non-acupoints group with statistical significance(P0.05),the activity of D1、D2 of model with acupoints and the expression of D1 mRNA、D2 mRNA were lower than the model with Naltrexone(P0.01).The activity and expression of D4 were no significant difference between the model group,the medel with acupoints group,the model with non-acupoints and the model with naltroxone group after treatment(P0.05).Conclusions:Acupuncture obstructed the provocation of heroin euphoria memory through decreasing the activity of the D1、D2、D3 and the expression of the D1 mRNA、D2 mRNA、D3 mRNA.The medel with acupoints group was more significant than the model with non-acupoints group in decreasing the activity of the D1、D2、D3 and the expression of the D1 mRNA、D2 mRNA、D3 mRNA.The medel with acupoints group was more significant than the model with naltroxone group in in decreasing the activity of the D1、D2 and the expression of the D1 mRNA、D2 mRNA.D4 didn't act on the way of acupuncture obstructing the provocation of euphoria memory.
