摘要 应用HPLC方法检测人血清中卡铂(Carboplatin)浓度.采用C18分析柱及预柱,流动相为重蒸水,流速1.1ml/min,检测波长为229nm,并用乙醚和氯仿去除内源性杂质干扰。实验结果表明:卡铂与内源性杂质分离良好,保留时间为8.70~9.31min,在血清中卡铂回收率96.49%。卡铂在lμg/ml以上线性关系良好,相关系数r为0.9999,回归方程Y=0.373lX-0.1025,日内及日间变异系数均小于6%(n=5),最低检测浓度为1μg/ml。 Determination carboplatin in human serum by HPLC was reported. An human serum that has been eliminated internal foreign substance and protein using ether and chlorform was subjected to high performace liquid chromatography on a C18 stationary phase as both analytical and preparing column,the mobile phase with duplication evaporating water,at a flow rate of 1.1ml/min,and was detected at 229nm.The determination results: Carboplatin was separeted well with internal impurity, the retain time 8.70~9. 31min,and the rate of recovery 96.49 % in human serum,the limit of determination was l μg/ml.The linear relation of carboplatin when concentration was higher than 1ug/ml has been got,the equation Y =0.3731X-0.1025(r=0.9999). The RSD within-day and between-day were lower than 6 % (n = 5).
