摘要 目的:研究肿瘤坏死因子-α(TNF-α)在肠源性内毒素血症中的作用及丹参防治肝损伤机制。方法:用硫代乙酰胺(TAA)诱发大鼠急性肝损伤,检测血浆内毒素、TNF-α及丙氨酸氨基转移酶(ALT)水平变化,并与丹参治疗组进行比较。结果:TAA诱发急性肝损伤大鼠血浆内毒素及TNF-α含量升高,二者呈正相关;血浆ALT亦升高,与TNF-α亦呈正相关。丹参治疗组大鼠血浆TNF-α及ALT水平均低于肝损伤对照组。结论:TAA引起肠源性内毒素血症,内毒素促进TNF-α释放,TNF-α进一步介导肝损伤。 AIM:To study the effects of tumor necrosis factor-αon gut-derived endotoxemia in rats and the preventive and therapeutive effects of Radix Salviac Miltiorrhizac (RSM). METHODS: Endotoxin, TNF-α and ALT (alanine aminotransferase) levels were determined in plasma of acute hepatic injury rats induced by TAA (thioacetamide), the results were compared with that obtained from RSM group. RESULTS:Plasma endotoxin and TNF-α levels were increased in rats of TAA group and there were a positive correlation between endotoxin and TNF-α contents ( r =0.8326, P<0.05 ).Plasma ALT contents also increased in TAA group and correlated with TNF-α level( r=0.8237, P<0.05 ).RSM decreased plasma TNF-α and ALT contents at the same time. CONCLUSION:TNF-α might played an important role in endotoxin-mediated hepatic injury. RSM may displayed its preventive and therapeutic effects by way of TNF-α level down-regulation.
