摘要
目的观察增殖细胞核抗原在庆大霉素诱发的中毒性急性肾小管坏死动物模型中的表达并探讨其意义。方法成年Ｗｉｓｔａｒ大鼠皮下注射庆大霉素（４００ｍｇ／ｋｇ／ｄａｙ）２ｄ，对照组用ＮＳ。用免疫组化方法检测不同时相点肾组织中增殖细胞核抗原的分布及强度。结果在使用庆大霉素后的表达第５天明显增强，第１１天最强，至第１５天恢复正常。结论在细胞修复过程中对ＤＮＡ复制起重要的调节作用。它的表达可以作为肾小管上皮细胞修复的标志之一。
Objective To observe the expression of proliferating
cell nuclear antigen (PCNA) in the aminal model of gentamicin-induced toxic acute tubular
necrosis and approach its significance.Methods Adult Wistar rats received subcutaneous
injection of gentamicin (400mg/kg/day) for 2 days, while control animals received physiologic
saline. The distribution and intensity of PCNA were examined immunohistochemically.Result
After the gentamicin injection PCNA increased significantly at day 5, reached the peak at day 11
and returned to normal at day 15.Conclusion PCNA played important regulating role in DNA
replication during cell repair. The expression of PCNA may act as a maker for repair of renal
tubular epithelial cells.
