参附注射液预处理对犬心肌缺血-再灌注损伤的保护作用 被引量:12

摘要 目的 探讨参附注射液(SF)预处理对犬心肌缺血-再灌注损伤(MI/RI)的保护作用.方法 20只健康成年杂种犬随机分为低剂量SF组(2 ml/kg,SF2组)、高剂量SF组(4 ml/kg,SF4组)、生理盐水模型组(N组)和假手术组(S组),每组5只.开胸后结扎犬冠状动脉左前降支,建立在体急性心肌I-R模型.用放射免疫法检测术前(T1)、冠脉结扎前(T2)、再灌注前(T3)、再灌注1 h(T4)和再灌注2 h(T5)血清肌钙蛋白T(cTnT)含量,用免疫组化法检测T5心肌组织核因子(NF)-κB和热休克蛋白70(HSP70)的光密度值.结果 与S组比较,SF2、SF4和N组心肌HSP70和NF-κB、T2~T5时血清cTnT的含量显著升高(P〈0.05).与N组比较,SF2、SF4组血清cTnT的含量有所降低,心肌HSP70的光密度值高,NF-κB的光密度值减低(P〈0.05).结论 SF预处理可能通过降低血清中的cTnT的含量、增强心肌HSP70的表达和减低NF-κB的活性而对I-R损伤心肌产生保护作用. Objective To study the protective effects of shenfu injection(SF) on myocardial ischemia-reperfusion(I-R) injury in dogs. Methods Twenty adult healthy cross bred dogs were randomized into four grouds of SF 2 ml/kg group(SFz ), SF 4 ml/kg group(SF4 ), sham operated group(S) and normal saline(NS) control group(N), with 5 dogs each SF or NS were injected before operation. The myocardial I-R injury model was established by ligating the left anterior descending coronary artery. The contents of serum eadiac troponin T (cTnT) were measured before operation and ligation,at 30 rain after ligation, 1 h and 2 h after referfusion by radioimmunity. The expressions of heat shock protein 70(HSP70) and nuclear factor-KB(NF-KB) in the cardiac muscle tissues were measured by immunohistochemistry at 2 h after referfusion. Results Compared with group S, serum contents of cTnT, HSP70 and NF-KB of the other three froups were significantly higher (P〈0.05). Compared with group N, serum levels of cTnT were lower, the expression of HSP70 was increased and the expression of NF^B was lower (P〈0.05). Conclusion SF has a protective effect on the myocardial I-R injury, which may be related to its function of decreasing the levels of eTnT, and decreasing the expression of HSP70, and lowering the activity of NF-kB.
