顺铂致体外培养大鼠窦前卵泡损伤模型的建立

摘要 目的探索建立顺铂(cisplatin,CDDP)大鼠窦前卵泡损伤的模型。方法以SD大鼠窦前卵泡为研究对象,将5组不同剂量(0、0.25、0.5、1、2μg/mL)的CDDP加入到体外培养的大鼠窦前卵泡培养液中。作用24h后,用光镜和Hoechst33342/PI荧光双染观察窦前卵泡的形态学变化,用化学发光法检测窦前卵泡培养液中的雌二醇(E2)水平。结果 CDDP浓度≥1μg/mL时,窦前卵泡的形态发生明显改变,培养液中E2浓度降低,与空白对照组比较差异有统计学意义(P<0.01)。结论 1μg/mL的CDDP作用24h后即可明显抑制大鼠窦前卵泡的生长,出现闭锁现象。该模型的窦前卵泡形态及E2水平出现明显变化,与人类CDDP导致的卵巢功能早衰的病变过程相似。
