荧光双染和三维重建技术对蠕形螨的形态观察和功能探讨 被引量:3

摘要 目的分别采用两种荧光染料对人体蠕形螨染色后置于激光共聚焦显微镜下进行三维重建,对其重要结构进行功能性探讨。方法先用异硫氰酸荧光素(fluorescein isothiocyanate,FITC)对虫体进性初染,再用碘化丙啶(propidium iodide,PI)复染,置于激光共聚焦显微镜下,用Imaris5.0软件进行三维重建,观测、鉴定毛囊蠕形螨形态特征。结果异硫氰酸和碘化丙啶均能与虫体充分结合,在三维重建技术下清楚观察到了虫体外部形态及前段摄食通道等内部结构。结论荧光双染和三维重建技术的结合,对深入研究人体蠕形外部形态、内部结构及探讨其功能、意义奠定了坚实基础。 The aim of the present study is to reconstruct the three-dimensional (3D) image of the double stained Dernode:r fol- liculorum by using confocal laser scanning microscope (CLSM), and to study the function of its primary structures thereby. The speci- mens were initially stained with fluorescein isothiocyanate (FITC) and then double stained with propidium iodide (PI). Placing the double stained specimens under the CLSM, a 3D image was reconstructed by Imaris 5.0, by which observation and evaluation of the morphological characteristics of the Demoded folliculorum were conducted. Results showed that the specimens were well combined with FITC and PI. A clear image of the miteg external appearance and the anterior feeding canal were exposed by the high definition 3D pictures. Ifs indicated that the combination of fluorescent double stain and 3I〉reconstruction technique is fundamental on the further study of the mite's external morphology as well as its internal structures and functions.
