摘要
【目的】研究姜黄素对胃癌SGC7901细胞的生长抑制作用,并探讨其可能的作用机制。【方法】MTT比色法测定细胞增殖抑制率;Hoechst33342荧光染色及电镜观察细胞形态及结构变化;免疫细胞化学染色法观察survivin蛋白表达的改变。【结果】一定浓度姜黄素作用SGC7901细胞后,细胞生长受到明显抑制,且抑制率呈浓度和时间依赖性;荧光显微镜下可见核浓缩,聚集,碎裂等典型的凋亡特征;电镜下可见到凋亡小体形成;免疫细胞化学染色法显示随着姜黄素作用浓度增加,survivin蛋白表达降低。【结论】姜黄素在一定浓度范围内可以明显抑制胃癌SGC7901细胞增殖,并下调抗凋亡基因survivin的表达。
[Objective] To investigate the effect of curcumin on inhibiting the growth of gastric cancer SGC7901 cells and its possible mechanism. [Methods] Human gastric cancer SC-C7901 cell was co-cultured with curcumin in different concentrations. MTT assay was used to measure the level of the proliferation of SGC7901 cells. The morphological changes of cells were observed by fluorescence microscope' and transmission electron microscope. The expression of survivin was measured with immunocytochemical staining. [Results] A certain concentration of curcumin could obviously inhibit the proliferation of SGC7901 cells in a dose and time dependent manner. Some apoptotic features such as condensed, aggregated and broken nuclei could be seen under fluorescence microscope and TEM. Apoptotic body could be seen under electron microscope. Immunocytochemical staining showed that the expression of surviving protein decreased with the increase of the concentration of curcumin. [Conclusion] A certain range of concentration of cureumin can obviously inhibit the proliferation of human gastric cancer SGC7901 cells and down regulate the expression of survivin.
