摘要
【目的】探讨阿仑膦酸钠对体外培养大鼠成骨细胞(OB)增殖、分化及矿化的影响。【方法]用混合醇消化法分离培养新生SD大鼠颅骨0B,观察不同浓度阿仑膦酸钠(1×10^-10、1×10^-9、1×10^-8、1×10^-7、1×10^-6mol/L)对成骨细胞增殖与矿化的影响。采用倒置相差显微镜观察0B的形态,用噻唑蓝(MTT)法检测OB增殖情况,茜素红染色法观察0B矿化结节的形成。【结果]1×10^-6、1×10^-7、1×10^-8mol/L的阿仑膦酸钠均能促进成骨细胞的增殖;阿仑膦酸钠浓度为1×10^-8mol/L时成骨细胞ALP活力最强;1×10^-6mol/L对骨基质矿化小鲒有抑制作用,1×10^-8mol/L促进骨基质矿化功能。【结论】适宜浓度的阿仑膦酸钠促进成骨细胞增殖及分化,促进骨基质矿化结节生成。
[Objective]To explore the effect o.f alendronate sodium on proliferation and mineralization of osteoblast(OB) in rats. [Methods]The effect of different concentration(1 ×10^-10 , 1 ×10^- , 1 ×10^-8 , 1 ×10^-7 , 1 ×10^-6 mol/L)of alendronate sodium on proliferation and mineralization of OB were observed by using mixed enzyme digestion for isolating and culturing cranial OB in neonatal SD rats. The morphology of OB was observed under inverted phase contrast microscope. The proliferation was detected by MTT and the generation of mineralized nodules was observed by alizarin red staining. [Results] The 1 ×10^-6 , 1 ×10^-7 and 1 ×10^-8 mol/L alendronate sodium could facilitate the proliferation of OB, while 1 ×10^-8 mol/L alendronate sodium had the strongest motility of ALP in OB and could promote the mineralization of bone matrix, and 1 ×10^-6 mol/L alendronate sodium could inhibit the mineralized nodules in bone matrix. [Conclusion] The suitable concentra- tion of alendronate sodium can facilitate the proliferation and differentiation of OB, and promote the generation of mineralized nodules in bone matrix.
