二磷酸盐对成骨细胞增殖及分化成熟作用的实验研究 被引量:6

摘要 目的研究二磷酸盐类药物—阿伦磷酸对成骨细胞增殖及分化成熟的作用。方法将人成骨样MG-63细胞体外培养、传代后分为5组:空白对照组、阳性对照组(加入10-8M地塞米松)、3个实验组(分别加入10-6M、10-8M、10-10M阿伦磷酸)。培养数日后,行细胞增殖、碱性磷酸酶(ALP)活性、骨形态发生蛋白-(2BMP-2)、骨钙素(OC)、I型胶原(Coll.I)基因表达等检测。结果加入阿伦磷酸的3个实验组MG-63细胞数目较对照组显著增加,其峰值出现在浓度为10-8M时;同时,阿伦磷酸使ALP活性增强,而且BMP-2、OC、Coll.I等成骨细胞相关基因的表达增加。结论阿伦磷酸具有促进成骨细胞增殖及分化成熟,诱导骨形成的能力。 Objective To investigate the effect of alendronate on proliferation and osteogenic differentiation in MG-63 cells. Methods MG-63 cells were divided into five groups: negative control (medium alone), positive control(treated with 10^-8M dexamethasone), three experimental groups ( treated with 10^-6M, 10^-8M, 10^-10M alendronate respectively). After a few days, we analyzed the outcome. Cell proliferation was determined by MTT; Osteogenic differentiation of BMSC was evaluated by alkaline phosphatase bioassay; Gene expression assesses was by RT-PCR. Results alendronate significantly increased the cell number over controls, attaining peak levels at a concentration of 10^-8M. Alkaline phosphatase activity was increased. C, ene expression of BMP-2, Type I collagen and osteocalein was enhanced. Conclusion Alendronate, aside from its role as inhibitor of ostceclastic bone resorption, is promoter of osteoblast proliferation and maturation.
