摘要
建立并优化非抗凝猪血提取血红素的工艺。以非抗凝猪血为原料,经匀浆、超声波溶血、1%盐酸丙酮分离提取血红素,丙酮蒸馏回收后血红素浓缩析出,再经0.1mol/LNaOH溶液纯化即得血红素;利用紫外-可见吸收光谱与红外光谱对其进行定性检测,利用紫外分光光度法对其进行定量检测并计算纯度;选择匀浆时间、匀浆后双蒸水加入量、盐酸丙酮溶液与溶血液的体积比(V盐酸丙酮∶V溶血液)、搅拌抽提时间等4个因素对血红素产率的影响进行研究,以优化提取工艺。建立非抗凝猪血提取血红素的方法,平均产量为10.6g/kg非抗凝血,平均产率为1.06%,纯度达90.8%;确定的最佳提取工艺条件是:匀浆5min,匀浆后加入等体积双蒸水,V盐酸丙酮∶V溶血液=5∶1,搅拌抽提10min。利用非抗凝猪血提取血红素切实可行,为充分利用我国丰富的猪血资源提供了切实可行的技术路线。
To establish and optimize the extraction technology of heme from non-anticoagulant porcine blood.Non-anticoagulant porcine blood as the raw material was homogenated and haemolysised by ultrasonic wave.Heme was separated and extracted by 1 % hydrochloric acid acetone solution.Heme was concentrated and precipitated after acetone was distilled and reclaimed,then it was purified by 0.1 mol/L NaOH solution.Ultraviolet-visible absorption spectrum and infrared spectrum were applied for qualitative test,and ultraviolet spectrophotometry was utilized for quantitative detection of heme,and then the purity quotient was calculated.4 factors of homogenate time,the amount of double-distilled water,the volume ratio of hydrochloric acid acetone solutionand to hemolysate (Vhydrochloric acid acetone solution∶Vhemolysate)and agitating/extracting time were chosen to study the impact of them on yield of heme and to optimize the extraction technology.Extraction method of heme from non-anticoagulant porcine blood was established.The average output was 10.6 g per kilogram non-anticoagulant porcine blood,the average yield was 1.06 % and the purity quotient was 90.8 % .The determined optimum conditions of heme extraction technology were that homogenating for 5 min,adding equal volume double-distilled water,Vhydrochloric acidacetone solution∶Vhemolysate at 5∶1 and agitating/extracting for 10 min.It is practicable to extract heme from non-anticoagulant porcine blood and provides a practical technical route for taking full advantage of the rich porcine blood resources in our country.
