Sulfotransferase (ST) enzymes catalyze the sulfate conjugation of many hormones,neurotransmitters,drugs and xenobiotic compounds.In order to study molecular biology of rat kidney ST,cDNA library of rat kidney in pcDNA1vector was synthesized by Gubler's method.Then,the library was screened with labled oligonucleotide as probe,and consequently,four positive clones were selected as possible sulfotransferase.One of them was sequenced by the dideoxyl chain termination method.It showed that the full length cDNA fragment(namely STK 63 )coutained 1277 bp and deduced amino acid of 296,which shared 84% amino acid identity with a human liver sulfotransferase in the GenBank TM/EMBL Data Bank with acceession number U66036 prot.Northern blot anlysis was performed with RNA samples of kidney,heart,liver,muscle,stomach,lung,intestine,spleen,testis and brain.A band hybridized with STK 63 cDNA fragment was detected at about 1.3 kilobase in kidney RNA sample,but no hybridization band was observed in other tissues.It implied that STK 63 cloning might be a specfic rat kidney sulfotransferase.
