摘要
采用细胞悬浮培养的方法,以烟草红花大金元细胞为材料,研究了几种有机营养物质、辅酶Q10(CoQ10)合成前体物质及Co60-γ射线对烟草悬浮培养细胞生长和辅酶Q10形成的影响实验证明,肌醇、酵母膏、盐酸硫胺素对烟草悬浮培养细胞生长和CoQ10含量都有一定的影响当ρ(肌醇)=100mg/L时,每瓶CoQ10比对照增加了90μg当ρ(盐酸硫胺素)=4mg/L时,细胞干质量、CoQ10含量及总量均随其浓度增加而升高当w(酶母膏)=0.5%时,细胞干质量、CoQ10含量和CoQ10总量均最高在CoQ10生物合成的前体中,甲硫氨酸(Met)对细胞生长无明显作用,但能促进CoQ10的形成实验还证明,Co60-γ射线辐射处理时,052C/kg剂量处理可促进1~3次继代培养细胞的生长;
With suspension cultured cells of tobacco, the effect of organic nutrient, precursors of CoQ 10 and Co 60 γ-ray irradiation on cell growth and CoQ 10 formation was studied. It showed that myoinositoland yeast extract and thiamine hydrochloride had effect on cell growth and content of CoQ 10 . When treated with 100 mg/L of myoinositoland (volumic mass), the dry weight of cells by was increased by 1.0 over the control; When treated with 200 mg/L of myoinositoland, the total content of CoQ 10 in each bottle was increased by 90 μg over the control. When treated with thiamine hydrochloride, the dry weight of cell, the conternt and total amount of CoQ 10 were increased. When treated with 0.15% yeast extract (mass fraction), the dry weight of cell, the content and total amount of CoQ 10 were all in the highest level. Met, the precursor of CoQ 10 , could promote the formation of CoQ 10 , but had no effect on cell growth. Effect of Co 60 γ ray irradiation on the cell growth and formation of CoQ 10 were also studied.
