同型半胱氨酸对ECV-304细胞NF-κB活性的影响及金雀异黄酮的保护机制

摘要 目的检测同型半胱氨酸(HCY)对人脐静脉内皮细胞株ECV-304的损伤对细胞核因子(NF-κB)活化的影响及金雀异黄酮(GEN)的保护作用。方法以5.0mmol/L的HCY作用于ECV-304细胞:设不同浓度的GEN(10、50、100μmol/L)保护组孵育12h后,再加入5.0mmol/L的HCY,通过四噻氮唑盐(MTT)比色法观察细胞活性,通过Western印迹法、免疫组化观察NF-κB表达情况。结果 HCY可降低ECV-304细胞的细胞活力,NF-κB表达增强;而在加入不同浓度的GEN预处理后,细胞活性显著增高,NF-κB表达水平显著下降,且呈现剂量依赖性(r=-0.95,P<0.05)。结论 HCY可作用于ECV-304细胞,活力下降,上调NF-κB蛋白及其靶基因的表达,而GEN能逆转HCY所引起的NF-κB活性增高的作用。 Objective To study the effect of homocysteine(HCY)on activation of NF-κB in ECV-304 and explore the protection of Genistein(GEN).Methods MTT assay was used to detect cell viability,and the expression of NF-κB was observed by Western blot and immunohistochemistry.Results The cellular viability of 5.0 mmol/L HCY 〔(41.27±5.09)%〕 was lower than those of the other groups,and the expression of NF-κB increased too.After co-treatment of 5.0 mmol/L HCY and different concentration GEN(10,50,100 μmol/L),the cellular viability of each group had elevated and the expression of NF-κB decreased.Conclusions HCY can lead to endothelial dysfunction in ECV-304 by increasing the activity of NF-κB.GEN can protect the cell viability and decrease the expression of NF-κB.
