大豆异黄酮抗脂质过氧化作用及其机制初探 被引量:50

ANTI-LIPID PEROXIDATIVE ACTIONS OF SOYBEAN ISOFLAVONES AND THEIR MECHANISM

摘要 目的 :观察大豆异黄酮对脂质过氧化作用的特点并探索其机制。方法 :在亚铁离子(Fe2 + )诱导亚油酸脂质过氧化体系中用电子自旋共振波谱法和比色法检测体系中脂质自由基、硫代巴比妥酸反应物质 (TBARS)和共轭双烯水平变化。结果 :无论在脂质过氧化启动前还是启动后向体系中加入 5~ 2 0 0 μmol大豆异黄酮均显著抑制TBARS的生成 ,但对体系中共轭双烯水平无影响。大豆异黄酮的作用特点类拟α -维生素E ,只是作用强度弱一些。电子自旋共振波谱分析显示 ,5~ 2 0 0 μmol大豆异黄酮对脂质过氧化体系中产生的脂质自由基具有显著的清除效果 ,此作用随大豆异黄酮浓度升高而增强。大豆异黄酮与体系中诱导剂铁离子间未观察到螯合现象。结论 :大豆异黄酮清除脂质过氧化体系中的脂质自由基是抑制过氧化链式反应的重要途径。 Objective:The effects of soybean isoflavones(SI) on lipid peroxidation were studied in the system of linoleic acid peroxidation induced by Fe 2+ . Methods:The changes of lipid free radicals, thiobarbituric acid-reacting substances(TBARS) and conjugated dienes in the system were measured through electron spin resonance(ESR) and colorimetry. Results:The lipid peroxidation in the system was inhibited significantly by 5-200 μmol by addition of SI whether before initiation of lipid peroxidation or after. The inhibition mode of SI was exactly similar to α-tocopherol, only its action was less a bit. The analyzing results of ESR showed that lipid free radicals in the system were strongly scavenged by 5-200 μmol SI, and the scavenging action was enhanced with the increase of SI. In addition, there was no chelation between SI and Fe 2+ , an inductive ion for lipid peroxidation. Conclusion:We concluded that it was a main mechanism for SI to suppress the peroxidative chain reaction by scavenging lipid free radicals in lipid peroxidative system.
