摘要
目的初步探索纳米碳酸钙对大鼠精子和外周血淋巴细胞DNA的影响。方法将32只雄性Wistar大鼠按体重随机分为4组,分别为阴性对照组(生理盐水),低剂量组(0.8mg/ml纳米碳酸钙),中剂量组(4mg/ml纳米碳酸钙),高剂量组(20mg/ml纳米碳酸钙)和用气管注入法对动物进行染毒,每周染毒一次,连续五周。对外周血淋巴细胞DNA的损伤用彗星实验进行分析,用精子畸形试验观察畸形率。结果染毒期间各组动物体重均有增长,各组间的增长量与对照组比较,差异均无统计学意义(P>0.05);各染毒组精子畸形率均高于对照组,但是差异没有统计学意义(P>0.05);各染毒组外周血淋巴细胞DNA的迁移率均明显高于阴性对照组,中、高剂量组与阴性对照组之间的差异有统计学意义(P<0.05)。结论在本试验条件下,纳米碳酸钙可造成大鼠外周血淋巴细胞DNA的损伤。
Objective To explore primarily the effect of nano-CaCO3 on sperm and peripheral blood lymphocytes DNA of rats. Methods 32 male rats were randomly divided into negative control group and three experimental groups which were exposed to 0. 8mg /ml,4mg /ml and 20mg /ml nano-CaCO3 ,respectively. The rats were instilled with physiological saline and different doses of nano-CaCO3 once a week for five weeks through tracheal injection method. Comet assay was performed to detect peripheral blood lymphocytes DNA damages. Sperm abnormality test was used to observe the rate of sperm deformity. Results Each group animals body weight increased during exposure,but there was no significant difference in comparison with negative control group(P 0. 05); The rate of sperm deformity in three exposed groups was higher than that in the negative control group, but there was no significant difference among the four groups (P 0. 05). The results also showed that serious DNA damage of peripheral blood lymphocytes was induced by nano-CaCO3 at three exposed groups compared with the negative control group. The DNA damage of 4 mg /ml and 20 mg /ml nano-CaCO3 was significant higher than that of the negative control groups (P 0. 05). Conclusions Nano-CaCO3 can induce peripheral blood lymphocytes DNA damage.
