摘要
利用PCR扩增、克隆和测序的方法对青岛市红岛虾池生长的缘管浒苔(Enteromorpha linza)的5.8S rD-NA及其转录间隔区ITS序列进行了分析,并依据ITS序列构建了浒苔属不同种之间的进化树。结果表明,获得了核糖体DNA完整的ITS和5.8S rDNA序列,ITS1为195 bp,5.8S rDNA为155 bp,ITS2为184 bp,总长度GC含量为63%。缘管浒苔与浒苔和曲浒苔的序列差异百分率较小,分别为2.4%和4.6%,与(U.olivascens)的序列差异百分率最大,为16.7%。Kimura-2参数遗传距离结果表明缘管浒苔和浒苔的遗传进化关系较近。
Enteromorpha linza is sampled from the shrimp ponds at Hongdao of Qingdao.Its 5.8S rDNA and internal transcribed spacer sequence(ITS) is analyzed.The PCR technology is used for the sequence amplification,clone,and determination.A phylogenetic tree of the various species is set up from the ITS sequences.It is learnt from the results that a complete ITS and 5.8S rDNA sequence is obtained,including 195 bp of ITS1,155 bp of 5.8 SrDNA,and 184 bp of ITS2.Its GC content is 63% of the total seqnence.The difference between E.linza and E.prolifera is 2.4%,it is 4.6% between E.linza and E.flexuosa,and it is 16.7% between E.linza and U.olivascens,the maximum of the three.The Kimur-2 parameter reveals that the genetic relationship between E.linza and E.prolifera is closer.
