摘要
目的:检测原发性胰腺导管腺癌组织中P21ras、转化生长因子-α(TGF-α)及表皮生长因子受体(EGFR)蛋白的表达情况。方法:应用免疫组织化学SP法检测30例原发性胰腺导管腺癌和10例正常胰腺组织中P21ras、TGF-α及EGFR蛋白的表达情况。结果:原发性胰腺导管腺癌组织中P21ras、TGF-α及EGFR的阳性表达率分别为63.3%(19/30)、80.0%(24/30)和70.0%(21/30),均高于正常胰腺组织的1/10、2/10和0(P分别为0.008、0.001和<0.001);P21ras的表达与胰腺癌的病理分级(P=0.042)、临床分期(P=0.007)及淋巴结转移(P=0.023)关系密切,而TGF-α及EGFR的表达与上述指标无明显相关性(P>0.05);胰腺癌组织中TGF-α与EGFR的表达密切关联(rP=0.607,P<0.001)。结论:胰腺癌组织中P21ras蛋白表达是一早期而普遍的事件,检测P21ras蛋白有助于胰腺癌的早期诊断;TGF-α与EGFR能够形成自分泌环,参与胰腺癌的发生。
Aim:To investigate the expressions of P21ras,TGF-α and EGFR proteins in pancreatic carcinoma(PC) tissue.Methods:A total of 30 primary pancreatic ductal adenocarcinoma samples and 10 normal pancreatic tissue samples were collected to detect the expressions of P21ras,TGF-α and EGFR proteins by immunohistochemistry method.Results:The positive rates of P21ras,TGF-α and EGFR in PC tissue were 63.3%(19/30),80.0%(24/30) and 70.0%(21/30),respectively,higher than those of 1/10,2/10,and 0 in normal pancreatic tissue samples(P was 0.008,0.001,and 0.001).The expression of P21ras,but not TGF-α or EGFR,was related with histological grade(P=0.042),clinical stage(P=0.007) and lymph node metastasis(P=0.023) of PC tissue.Positive association was observed between the expressions of TGF-α and EGFR in PC tissue(rP=0.607,P0.001).Conclusion:Increased expression of P21ras is an early molecular event in the pathogenesis of PC,and its detection may be useful for early diagnosis. The autocrine loop of TGF-α/EGFR may contribute to the carcinogenesis of PC.
