摘要
目的通过2,4-二硝基氯本(DNCB)加乙醇、乙酸复合法建立大鼠UC模型,观察康肠宁对该模型的治疗作用,并探讨相关机制。方法采用DNCB+乙醇+乙酸法复制大鼠模型,模型成功后将大鼠随机分组,同时设立对照组。给药期间观测各组动物的精神、饮食、大便等情况,治疗结束后采血以免疫组化法测CD4、CD8及其比值,处死大鼠取结肠做病理学检查,刮取结肠黏膜以放免法测TNF-α。结果模型组大鼠可见稀便或软便,精神倦怠,活动、饮食明显减少,毛发欠光泽;结肠充血、水肿,溃疡形成,甚至肠壁变厚,肠腔变大;镜下炎性细胞浸润明显,个别可见肉芽肿,伴不同程度的纤维化;外周血CD4、CD8较正常组显著下降(P<0.01),组织TNF-α有所下降(P>0.05)。给予康肠宁中、高剂量组治疗后稀便渐停,饮食、活动逐渐恢复正常;结肠黏膜充血、水肿基本消失,个别可见愈合期溃疡,镜下炎性细胞明显减少,纤维化程度明显减轻;外周血CD4、CD8较模型组显著升高(P<0.05),组织TNF-α有一定程度升高(P>0.05)。结论康肠宁对DNCB+乙醇+乙酸法建立的大鼠UC模型有一定的治疗作用,其机制与调节免疫、清除炎症有关。
Objective Ulcerative colitis(UC)rat model was established by dinitrochlorobenzene(DNCB)plus ethanol and acetic acid,so as to observe the effects of Kangchangning on the model and to explore the relevant mechanism. Methods After the model was successfully established,rats were randomly divided into 3 groups,and the normal control group was also established.The mental status,diet and bowel movement of the animals were observed during the medication.After treatment,CD4,CD8 and CD4/CD8 were measured with immunohistochemical method.Pathological examination of colonic mucosa was carried out.Colonic mucosa was scraped for the detection of TNF-αwith radioimmunoassay.Results Soft feces,fatigue,lusterless hair,obviously reduced activity and diet were shown in the rats of the model group.The colons were seen with hyperemia,edema,ulcer formation,or even thickening colon walls and enlarging of the colonic cavity.Obvious infiltration of the inflammatory cells was found under microscope.In some serious cases,granuloma complicated with different degrees of fibrosis in the colons was presented.Peripheral blood CD4,CD8 decreased significantly compared to that of the normal group(P〈0.01).symptoms were relieved,pathological changes of colonic mucosa such as ulcers and inflammation were reduced,and peripheral blood CD4 and CD8 increased significantly compared to that of the model group(P〈0.05)in middle-and high-dose Kangchangning groups,but the content of TNF-αhad no significant changes(P〉0.05).Conclusion Kangchangning has certain therapeutic effect on the UC rat model established by DNCB plus ethanol and acetic acid,and its mechanism is concerned with immunoregulation and removal of inflammation.
