摘要
硒蛋白的组分分析的难题是硒氨基酸在水解中不稳定。Blau指出硒蛋氨酸(SeMet)在6mol/L HCl、110℃、7h水解中几乎全部被破坏,Shepherd也得到同样的结论。硒胱氨酸和硒半胱氨酸也有相似的行为。其后,前人曾提出各种方法对此加以改进,例如加入各种还原剂以及通氮除氧,目的是防止硒氨基酸被氧化成亚砜或砜。最近刘曼西等提出在4mol/LHCl、50℃,50h的水解方法,但未通氮除氧。
A modified hydrolysis method ( 4mol/L HC1,50℃,50h,in N2) for determining selenoprotein has been established.The recovery of this method is 84% with respect to the bound selenomethionine.The reasons for the comparatively high recovery of this method were found by experiments.This method would be of significance to the determination of selenoprotein.
