摘要
目的:测定杭白菊茎叶中蒙花苷、木犀草素的含量及建立其HPLC特征指纹图谱。方法:色谱柱:Waters Symmetry-Shield^(TM)RP_(18)(250 mm×4.6 mm,5μm);流动相:乙腈-甲醇(9:1)与0.1%磷酸水溶液梯度洗脱;检测波长:340 nm;流速:1.0ml·min^(-1);进样量:20μl。结果:蒙花苷与木犀草素分别在0.399~1.994μg和0.200~1.800μg线性关系良好,平均回收率分别为101.7%和98.3%,RSD均小于3.0%。13批杭白菊茎叶中蒙花苷和木犀草素的含量分别为(0.258±0.089)%、(0.094±0.056)%;HPLC特征指纹图谱中杭白菊主要成分均得到很好分离,获得13个共有特征峰。结论:本方法重复性好,专属性强,为杭白菊茎叶的质量控制和开发利用提供科学依据。
Objective: To determinate linarin and luteolin in the stem and leaf of Dendrantherna morifolium ( Ramat. ) Tzvel. and to establish its fingerprint. Method: The ethanol extract was separated on a Waters SymmetryShieldTM RPts column (250 mm × 4. 6 mm, 5 μm) , and gradiently elated using acetonitrile-methanol (9:1 ) and 0. 1% phosphoric acid solution as the mobile phase. The de- tected wavelength was set at 340 nm with a PDA detector. Flow rate was 1.0 ml·min-1 and 20 p.1 was injected. Result: The linear ran- ges of linarin and luteolin were 0. 399-1. 994 μg,0. 200-1. 800 μg and the average recoveries were 101.7% and 98. 3% ( RSD 〈 3. 0% ) respectively. The content of linarin and luteolin in the stem and leaf of Dendranthema morifolium ( Ramat. ) were (0. 258 + 0. 089) %, (0. 094 ± 0. 056) %, respectively. In the fingerprint analysis, 13 common peaks in the stem and leaf of Dendrantherna morifolium ( Ramat. ) Tzvel. were achieved. Conclusion: This method can be used scientifically to quality control of stem and leaf of Dendrantherna morifolium ( Ramat. ) Tzvel.
