基因芯片筛选口腔扁平苔藓差异表达基因的研究 被引量:2

摘要 目的用基因芯片技术筛选口腔扁平苔藓(orallichen planus,OLP)相关基因。方法分别从人OLP组织和正常口腔黏膜组织中提取总RNA并纯化为mRNA,再将2种组织的mRNA分别进行荧光标记,然后与含有32 050个位点的基因的人类基因芯片进行杂交,探针长度是60mer,杂交信号用Axon Genepix 400B MicroarrayScanner扫描仪扫描,结果用芯片图像分析软件(Genepix Pro V6.0和Genesring)进行分析。结果经过杂交筛选并与正常组织比较,从32 050个基因中筛选出差异有统计学意义的表达基因142个,其中上调的基因有25个,下调的基因有117个。结论 OLP的发生、发展过程中存在着多条不同功能基因表达调控的改变,运用基因表达谱芯片可以初步筛选出OLP相关基因。 Objective This study aims to screen the differentially expressed genes related to the pathogenesis of oral lichen planus(OLP) by gene array. Methods The total RNA was extracted from oral lichen planus tissues and normal oral tissues, and purified to mRNA. The mRNA was labelled with fluorescence,and then hybridized with the package that contains 32 050 genes of the human gene chip , the probe was 60 mer in length. The hybridzation signal was scanned with the Axon Genepix 400B Microarray Scanner, and the result was analysed with Genepix pro V6. 0 and Genesring software. Results There were 142 differentially expressed genes and 25 genes were up-regulated and 117 genes were down-regulated. Conclusion Many genes with different functions changed, which may play some roles in the progression of OLP, and Geneehip technology can be used for screening OLP-related genes preliminarily.
