依达拉奉对犬肾缺血再灌注损伤的保护作用 被引量:4

Effects of Edaravone on Renal Ischemia-Reperfusion Injury in Dogs

摘要 目的:探讨依达拉奉对犬急性肾缺血再灌注损伤的影响及相关机制。方法:24只成年雄性杂种犬,分为3组(n=8):假手术组(S组);缺血再灌注组(I/R组);治疗组(Edaravone组)。在肾缺血60 min后完全恢复灌流。术后犬存活3 d,术前及术后1,2,3 d检测血清肌酐(Cr)、尿素氮(BUN)浓度;检测肾组织超氧化物歧化酶(SOD)活性,丙二醛(MDA)及髓过氧化物酶(MPO)含量;原位末端脱氧核苷酸转移酶标记(TUNEL)法检测肾组织中凋亡细胞,计算细胞凋亡指数(AI);光镜下观察肾组织病理学改变。结果:Edaravone组与I/R组术后3天连续比较,血清Cr、BUN浓度有显著降低。Edaravone组与I/R组比较,SOD活性升高,MDA浓度降低,MPO下降,AI降低(P<0.05),病理损伤明显减轻。结论:依达拉奉能减轻肾缺血再灌注损伤,其机制可能与减轻肾脂质过氧化反应和抑制细胞凋亡,减少炎症有关。 Objective: To investigate the effects of edaravone on the acute renal ischemia/reperfusion(I/R) injury in dogs.Methods: Twenty-four adult male Mongrel dogs were randomly divided into three groups with 8 in each group: sham operation group,ischemia/reperfusion(I/R) group,edaravone treatment group.Blood samples were obtained for determination of blood creatinine(Cr) and urea nitrogen(BUN) concentrations before operation and 24,48,72 hours after operation.The dogs were killed on the third day after operation and left kidneys were removed for determination of SOD activity and MDA and MPO concentrations.The apoptosis in the nephridial tissue was assessed by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL),and apoptotic index(AI) was calculated.The changes of renal tissue were observed under microscope.Results: Blood Cr and BUN concentration in edaravone group decreased significantly than in I/R group in 3 days after operation.MDA and MPO concentrations were decreased,SOD activity was increased significantly and AI was decreased significantly in treatment group as compared with that respectively in I/R group on the 3rd day after operation(P〈0.05).Microscopic examination showed that the renal I/R injury was significantly attenuated by edaravone.Conclusion: Edaravone can attenuate renal damage induced by I/R through inhibiting oxidative stress,apoptosis and inflammation.
