摘要
目的 探讨大剂量甲基强的松龙对脊髓损伤后神经细胞凋亡的影响。 方法 将6 0只大鼠分为脊髓损伤后应用大剂量甲基强的松龙组 (A组 ,30只 )和单纯脊髓损伤组 (B组 ,30只 )。损伤后 1,3,7,14,2 8d ,对脊髓损伤区进行细胞凋亡的检测 (原位末端脱氧核糖核苷酸转移酶介导dUTP标记法 ,TUNEL)以及Bcl- 2蛋白表达的测定 (免疫组化SP法 )。 结果 A、B两组中均发现凋亡细胞及Bcl- 2蛋白阳性表达。大鼠脊髓损伤后 3d图像分析表明 ,细胞凋亡率分别为 :A组 (12 .43± 5 .16 ) ,B组 (36 .2 7± 6 .2 8) ;Bcl- 2蛋白阳性细胞分别为 :A组 (37.6 8± 5 .83) ,B组 (2 1.48± 7.83)。两组比较 ,差异有显著性意义 (P <0 .0 1和 0 .0 5 )。 结论 脊髓损伤可导致神经细胞凋亡 ,大剂量甲基强的松龙能抑制细胞凋亡。
Objective To investigate the effect of high dose methylprednisolone(MP) on cell apoptosis after spinal cord injury (SCI). Methods Sixty experimental rats were divided equally into 2 groups, Allen's weight drop spinal cord injury and high dose methylprednisolone group (Group A) and Allen's weight drop injury group (Group B). The cell apoptosis of spinal slice from the injured spinal cord was examined by the terminal deoxynucleotidal transferase mediated DUTP biotin nick end labeling (TUNEL) reaction, and the expression of Bcl 2 measured immunohistochemically 1, 3, 7, 14 and 28 days post operatively. The positive cells were quantitated by a computer image analysis system. Results Positive expression of apoptotic cells and Bcl 2 protein appeared both in Group A and Group B. SCI for 3 days, the computer image analysis system showed the apoptosis rate was 12.43 ± 5.16 in Group A and 36.27±6.28 in Group B; Bcl 2 protein expression positive cells were 37.68±5.83 in Group A and 21.48 ± 7.83 in Group B. There was a significant statistic difference remained between Group A and Group B ( P < 0.01 and 0.05 ). Conclusions The high dose methylprednisolone can inhibit apoptosis after SCI.
