摘要
目的:由于目前保健品中核黄素检测常用的荧光分光光度法操作繁琐、精密度较差,所以建立并优化适用于复合维生素保健品核黄素含量检测的HPLC-荧光检测方法,并与荧光分光光度法进行比较。方法:建立使用Diamonsil C18色谱柱(150×4.6 mm,5μm),以1%醋酸溶液-甲醇(70:30)为流动相,以荧光检测器(激发波长:445 nm,发射波长:530 nm)进行检测的液相色谱方法。对七种保健品中的核黄素分别用两种方法测定,考察结果之间的差别。结论:HPLC法结果的精密度和标准曲线线性明显优于荧光分光光度法,且操作更为简便,更适于保健品中核黄素的测定。
Objective:To develop a method for determination of riboflavin in health protection products by HPLC with fluorimetric detection,and to compare with the fluorescence spectrophotometry method.Methods:The chromatographic conditions included a Diamonsil C18 column(150×4.6 mm,5 μm) with the mobile phase of methanol-1% acetic acid water(30:70,v/v) and fluorimetric detector at a wave 1ength of 445 nm for excitation and 530 nm for emission.Samples were analyzed by two methods.Conclusion:The HPLC method is more suitable for determination of riboflavin in health protection products.
