摘要
目的研究高糖环境对体外培养的大鼠海马神经元乏氧损伤时凋亡的影响以及脑神康胶囊对损伤神经元的保护作用。方法原代培养大鼠海马神经元,将细胞随机分成8组:对照组、高糖组、黄嘌呤(X)/黄嘌呤氧化酶(XO)组、川芎嗪组、生理盐水组、脑神康低、中、高剂量组。干预后测细胞存活率及凋亡率,采用Real-time PCR法检测各组海马神经元HIF-1α mRNA、Bcl-2 mRNA和Bax mRNA的表达及Bc-2/Bax比值。结果与对照组相比,高糖组及X/XO组神经元存活率、Bcl-2 mRNA表达水平和Bcl-2/Bax的水平均显著下降,神经元凋亡率、HIF-1α mRNA和Bax mRNA表达水平均显著升高(均P<0.01);与X/XO组比较,川芎嗪组及脑神康各浓度组神经元存活率、Bcl-2 mRNA表达和Bcl-2/Bax的水平均明显上升,而神经元凋亡率、HIF-1α mRNA和Bax mRNA表达均明显下降。结论脑神康胶囊能够明显抑制高糖环境下大鼠海马神经元的凋亡,对高糖环境下海马神经元的乏氧损伤有保护作用,其作用机制之一是下调HIF-1α mRNA表达、上调Bcl-2 mRNA表达、改善乏氧环境及抗细胞凋亡。
Objective To research the effect of Naoshenkang capsule on expression of hypoxia-inducible factor-1 alpha (HIF-1α) of oxidative damaged rat hippocampal neurons in high glucose. Methods The primarily cultured neonate rat hippocampal neurons were randomly divided into eight groups: control,high glucose,xanthine/xanthine oxidase (x/xo),ligustrazine,normal sodium and the small,middle,and large dosage of Naoshenkang groups. The rate of cell survival and apoptosis were measured,and the method of real-time PCR was used to detect the mRNA expression of HIF-1α,Bcl-2,Bax. Results The survival rate,the expression of Bcl-2 mRNA and the ratios of Bcl-2 and Bax in the high glucose group were lower,but apoptosis and the expression of HIF-lα and Bax mRNA higher than those of x/xo group (P0.01). The survival rate,the expression of Bcl-2 mRNA and the ratios of Bcl-2 and Bax in the ligustrazine group and all Naoshenkang groups were significantly higher,but apoptosis and the expression of HIF-lα and Bax mRNA lower than those of x/xo group. Conclusions Naoshenkang capsule can significantly inhibit the apoptosis of rat hippocampal neurons in the high glucose,and make a protective effect on hypoxia injury. Its mechanism may be that Naoshenkang capsule can down-regulate HIF-lα mRNA expression,up-regulate Bcl-2 mRNA expression,improve the hypoxia and resist apoptosis.
