奥沙利铂对肺腺癌A549细胞株的放疗增敏作用 被引量:2

摘要 目的观察奥沙利铂(L—OHP)对体外培养非小细胞肺癌A549株的放疗增敏作用。方法体外培养非小细胞肺癌株A549,分为放疗+药物组、单纯药物组、单纯放疗组和对照组。采用MTT法观察相同L.OHP药物浓度(≤IC10)、不同放射剂量(1Gy、1.4Gy、1.8Gy、2Gy、3Gy)及不同药物浓度(0.488mg/mL、0.244mg/mL、0.122mg/mL、0.06lmg/mL、0.031mg/mL)、相同放射剂量(2Gy)对A549细胞株的抑制作用。计算细胞抑制率和增敏比,应用SPSS13.0处理数据,分析各因素对人肺癌细胞A549的抑制作用。结果MTT检测显示L—OHP浓度为0.488mg/mL为最佳实验浓度。非小细胞肺癌A549细胞生长抑制率与放射剂量、药物浓度呈正相关。流式细胞仪分析结果显示各处理组细胞在S期时的比例有显著性差异(P〈0.05)。结论使用L—OHP可增强放射线对非小细胞肺癌细胞的作用。 Objective To explore the enhancing radiosensitivity of oxaliplatin on human Non-small Lung Cancer cell A549. Methods The Non-small Lung Cancer cell A549 were put into four groups randomly .One group was effected by oxaliplatin with same dose (~〈IC^0) and radiation with different dosez (1Gy, 1.4Gy, 1.8Gy, 2Gy, 3Gy), or the different oxaliplatin doses (0.488mg/mL, 0.244mg/mL, 0.122mg/mL, 0.061 mg/mL, 0.031mg/mL) and same dose radiation (2Gy). The two of the group were effected only by radiation or oxaliplatin. Observing the effect of two group with MMT method, calculating the suppression rates and SER (sensitization enhancement ratio). Statistical analysis was performed by ANOVA test, multiple correlation with the software SPSS13.0. Results The optimal dose of L-OHP was 0.488mg/mL by MTT method.There was significant differences among two group, showed positive correlation with the radiation dose and radiation time. Conclusions Oxaliplatin can enhance the radiosensitivity of Non-small Lung Cancer cell lines A549.
