摘要
AFLP是一种以PCR为基础的DNA指纹图谱技术。利用AFLP技术对从国际半干旱所(I-CRISAT)引进的9份花生抗旱品种绘制指纹图谱,通过引物E—ACA和与之匹配的MCAG和M—CAT,在300~6000bp的范围内共获得1577条AFLP扩增产物,每个品种有主带和次带至少71条之多,其中10条为多态性的带纹。
Amplified fragment length polymorphism(AFLP) is a novel PCR based DNA
fingerprinting technique. With the combinations of EACA primer paired with MCAG and MCAT,
respectively. Productions of 1577 segregating AFLP amplification in 9 groundnut (Arachis
hypogeae L.)varieties from ICRISAT(International Crops Research Institute for the SemiArid
Tropics)were identified. AFLP results generated more than 71 polymoiphic bands, contained
some major bands and some minor bands, ranging 3006000 base pairs, from one pair of EcoR I
primer and Mse I primer and up to 10 polymorphic bands can be detected with a single primer
for each varieties on the denatured gel.
