摘要
将体外重组的日本血吸虫中国大陆株28ku谷胱甘肽S转移酶(rSj28GST)与免疫刺激复合物(ISCOM)结合制备了rSj28GSTISCOM。用含85μgrSj28GST的rSj28GSTISCOM及100μgrSj28GST分别免疫BALB/c小白鼠,用常规ELISA检测体液免疫水平,以日本血吸虫中国大陆株尾蚴攻击后的减虫率表示免疫保护力。结果rSj28GSTISCOM免疫组的减虫率为25.5%,其荷虫数和空白对照组荷虫数相比有显著差异,而rSj28GST免疫组的减虫率仅为15.2%,与空白对照组相比差异不显著;ELISA检测结果,rSj28GSTISCOM免疫组比rSj28GST免疫组不仅抗体水平高。
The in vitro recombinant 28 ku Glutathione st transferase of schistosoma japonicum (rSj28 GST) was combined with immune stimulative complex (ISCOM) for preparing rSj28 GST ISCOM. Balb/c mice were immunized with rSj28 GST ISCOM containing 85 μg rSj28 GST and 100 μg rSj28 GST, respectively, and then the humoral immunity efficacies were measured by a routine ELISA, and the immune protections were represented with S.japonicum cercariae. The test results revealed that the worm reduction rate of mice immunized with rSj28 GST ISCOM was 25.2%, which was significantly higher than that of mice immunized with rSj28 GST. And the former's antibody levels were also much higher than that of latters' (15.2%), and persisted for a longer interval.
