摘要
目的:观察活血药川芎嗪、水蛭素、丹参酮ⅡA和凝血酶对高转移人巨细胞肺癌PGCL3(简称PGCL3)和低转移人肺腺癌PAa(简称PAa)细胞的粘附和侵袭的影响。方法:用不同浓度的川芎嗪、丹参酮ⅡA、水蛭素和凝血酶分别处理PGCL3和PAa细胞,观察PGCL3和PAa细胞单独或与血小板作用后对纤维粘连蛋白的粘附和Boyden小室的侵袭改变。结果:川芎嗪、水蛭素和凝血酶在不同程度上增加PGCL3细胞的粘附,而丹参酮ⅡA抑制这种粘附。川芎嗪、丹参酮ⅡA和水蛭素抑制PGCL3细胞对Boyden小室的侵袭,凝血酶明显促进这个过程。川芎嗪、丹参酮ⅡA、水蛭素抑制血小板与PGCL3和PAa细胞侵袭的协同作用,凝血酶对这个过程有促进作用。结论:活血药对肿瘤细胞的侵袭和转移可能既有抑制作用又有促进作用。
Objective: To investigate the influence of some blood activating drugs like Tetramethylpyrazine, Tanshinone ⅡA, Hirudin and thrombin on adhesive and invasive behavior of PGCL3 and PAa cell lines. Methods: Using the above mentioned blood activating drugs in various concentration to treat PGCL3 and PAa cells, and the changes in adhesion to fibronectin and invasion in Boyden Chamber of these cells, alone or after interacted with human platelets, were observed. Results: Tetramethylpyrazine, Hirudin and thrombin could increase the adhesion of cells to fibronectin and Tanshinone ⅡA decrease it. Tetramethylpyrazine, Tanshinone ⅡA and Hirudin inhibited the invasion of PGCL3 cells in Boyden Chamber, and thrombin augmented the process. Conclusion: The blood activating drugs may either inhibit or promote the invasion and metastasis of PGCL3 and PAa cells in the light of various conditions.
