褪黑素的镇痛作用和身体依赖性实验 被引量:14

摘要 本文在小鼠上观察褪黑素的镇痛作用和身体依赖性。热板法痛阈测试表明,腹腔注射(ip)褪黑素可产生显著的镇痛效应,并呈剂量依赖性。小鼠跳跃反应试验表明,在2d内7次皮下注射(sc)褪黑素或吗啡后,再ip纳洛酮,吗啡组小鼠出现显著的跳跃反应,而褪黑素组则无;小鼠竖尾反应表明,ip不同剂量褪黑素,小鼠都未出现竖尾反应。上述结果提示,褪黑素对小鼠无身体依赖性作用。 The analgesic effects and physical dependence of melatonin in mice were demonstrated. In the hot-plate test, it was observed that the mice treated with melatonin (ip, 20, 50, 80 mg·kg-1) showed a dose-related analgesic response. In the test of jumping-response in mice, the mice received 7 injections (sc) of melatonin or morphine, followed by naloxone 10 mg·kg-1(ip). Marked jumping was precipitated with naloxone after morphine, but not after melatonin (735 or 945 mg·kg-1). In mice, ip melatonin 45 or 90 or 180 mg·kg-1 exhibited a sedative effect, while ip morphine 15 mg·kg-1 caused a significant Straub's tail reaction and motor activity. These results indicate that melatonin has a significant analgesic effect without producing physical dependence in mice and it may be a new and effective analgesic drug.
