摘要
以加拿大美女樱的嫩茎为材料,进行了芽的生长与分化、不定芽的生根、试管苗的继代培养与留茬培养,以及试管苗的移栽、扦插和移植的研究,建立起快速繁殖技术。结果表明:MS+GA30.4mg/L+BA0.3mg/L+IAA0.2mg/L是培养芽生长的理想培养基;1/2MS+GA30.5mg/L+BA0.6mg/L+NAA0.1mg/L是生长芽分化培养的理想培养基;1/2MS+GA30.3mg/L+NAA0.1mg/L+BA0.5mg/L是不定芽分化继代培养的理想培养基;1/2MS+GA30.2mg/L+NAA0.1mg/L+IBA0.3mg/L是试管苗生根培养和生根继代培养的理想培养基;在温室苗床上,试管苗易移栽、扦插成活。移植的试管苗长势整齐而旺盛,根系发达。
Using Verbena canadensis Britt. as test material,the development of growing point,differentiation of adventitious buds,cutting and transplantation of tube seedlings for the tender stem of Verbena canadensis Britt. were studied,and the rapid propagation technology was established. The results showed that the ideal medium for development of growing point,differentiation of adventitious buds and regeneration was MS+GA3 0.4 mg/L+ BA 0.3 mg/L+IAA 0.2 mg/L,1/2 MS+GA3 0.5 mg/L+BA 0.6 mg/L+NAA 0.1 mg/L and 1/2 MS+GA3 0.3 mg/L+NAA 0.1 mg/L+BA 0.5 mg/L respectively. The optimum medium for growing and regeneration of roots was 1/2 MS+GA3 0.2 mg/L+NAA 0.1 mg/L+IBA 0.3 mg/L. The tube seedlings could be easily transplanted in seedling bed. The transplanted tube seedlings grew well with vigorous roots.
