放射治疗诱发肾癌细胞凋亡及机制探讨 被引量:1

Sudies on programmed cell death induced by ionizing irradiation and expression of P53 and Bcl 2 in Renal carcimona cell lines

摘要 目的探讨射线诱发肾癌细胞凋亡的作用和射线对肾癌细胞P53,Bcl-2基因表达的影响。方法利用末端脱氧核苷酰转移酶标记的流式细胞技术定性和定量分析射线诱导肾癌细胞凋亡。利用P53和Bcl-2单抗进行SPTM法免疫组织化学染色。结果0Gy、5Gy、10Gy、15Gy和20Gy照射后细胞凋亡量分别为1.02%、0.03%、7.03%、4.22%和36.09%。0Gy照射组P53表达标记指数为0.80%,20Gy照射后P53表达明显增加达23.80%。0Gy照射组Bcl-2基因表达为24.40%,20Gy照射后Bcl-2基因表达明显减少为1.80%。结论低剂量X射线照射诱发肾癌细胞凋亡量很低,20Gy时明显增加。射线诱发肾癌细胞凋亡伴随P53基因表达增加和Bcl-2基因表达减少,提示射线诱发细胞凋亡是多基因相互影响,相互作用的结果。 Objective\ To investigate the induction of apoptosis by X ray ionizing irradiation in Renal carcimona cell lines and the changes of X ray ionizing irradiation on the expression of P53 and Bcl 2 genes in Renal carcinoma cell lines. Method Immunohistological staining assay (SP TM assay) has been used for detect the changes of P53 and Bcl 2 gene expression after X ray ionizing irradiation. Results The percentages of apoptotic cells were 0.03%、7.03%、4.22% and 36.09% after ionizing irradiation of 5Gy、10Gy、15Gy and 20Gy respectively. Expression of P53 increased and Bcl 2 decreased significantly in 20Gy group. Conclution The percentages of apoptotic cells were very low with low dose radiation and increased with 20Gy radiation. The expression of P53 gene increased and Bcl 2 gene decreased greatly with 20Gy radiation than that of control group. It was indicated that the apoptosis induced by ionizing irradiation was regulated by some genes.
