摘要 目的研究EPO对心肌缺血再灌注损伤的作用及其机制。方法实验大鼠随机分为假手术组、模型组、EPO组,冠脉结扎和再松开复制动物模型,计算心律失常评分和心肌梗死面积,免疫组化法检测心肌组织EPOR、NF-κB、TNF-α的表达。结果与假手术组、模型组比较,EPO组经EPO预处理后心律失常评分和心肌梗死面积减少,EPOR的表达增强(P<0.01)NF-κB、TNF-α的表达显著减少(P<0.01)。结论 EPO对心肌缺血再灌注损伤有明显保护作用,其作用机制可能与上调EPOR表达及下调NF-κB的表达有关。 Objective To study the protective effect of EPOR in ischemia-reperfusion injured myocardium and the mechanism. Methods Rats were randomly divided into three groups: sham operation group, model group, EPO group. Experimental animal model was conducted by ligating and loosening left anterior descending coronary. Arrhythmia score, infarct size, and the expression of EPOR, NF-κB, TNF-αwere compared in the three groups. The expression of EPOR, NF-κB, TNF-α was determined by immunohistochemical staining. Results In EPO group, the arrhythmia score, infarct size, the expression of NF-κB and TNF-α were significantly decreased than that in sham operation group and model group(P〈0.01), while and the expression of EPOR was significantly increased.(P〈0.01).Conclusion EPO could protect myocardium from ischemia-reperfusion injury by up-regulating the expression of EPOR and down-regulating the expression of NF-κB.
