摘要
目的探讨麻疹野病毒的病原学特征,为WHO麻疹病毒参考资料库(databank)提供国内样品株,填补我省麻疹抗原分离的空白。方法采用组织培养法,选用敏感细胞分离抗原,并用中和试验法进行野毒株的鉴定。结果在B95-a细胞上共分离出10株麻疹野病毒,其中7份咽拭液同时用Vero细胞分离未获成功,但将10株分离株转种至Vero细胞上时,有4株出现麻疹的融合性细胞病变。1∶10稀释的麻疹单克隆抗体(12E)能中和所有分离株,在同期收集的9例麻疹患者血清中,4例麻疹IgM抗体阳性,但在1例麻疹IgG抗体滴度达1∶6400至1∶≥102400的患者中,也分离到了麻疹病毒。结论在湖南境内分离的10株麻疹野毒中,94—47株为目前国际上最大程度的麻疹变异株,具有一定抗体水平的儿童仍受到90年代野毒的攻击而形成临床感染。
Objective To probe the etiological characteristics of the
wild measles virus and to provide the information for the data bank of WHO.Methods The
tissue culture was used to select the sensitive cell to isolate the antigen.The neutralizatin test
was used to determine the wild strain.Results Ten strains of wild measles virus were isolated
from the B 95-a cell.Among these strains,7 samples of throat swab inoculated onto Vero cells
failed to grow any trace of measles virus when these 10 wild virus strains were inoculated onto
Vero cells, 4 of them showed confluent cytopathic effects(cpe).All the isolated strains could be
neutralized by the anti-measles hemoglobulin monoclonal antibody in 1∶10
titer(12E).Meanwhile 9 sera samples of measles patients were collected and the sera Igm of 4
samples showed to be positive.The measles virus was isolated from the sera of a patient with
the IgG titer reaching 1∶640 to 1∶>102400.Conclusion Among these 10 strains of wild
measles virus isolated in Hunan province,94-47 strains is the measles mutation strains with
the biggest degree internationally.The children with the antibody will still be infected by the 90's
wild virus. [
