摘要
观察术后不同时间内自体移植静脉增生内膜中移行白细胞与内膜增生的量效关系。方法:将24只家兔分为服药组与对照组,在股静脉移植至股动脉模型上,应用白细胞共同抗原CD_(45)免疫组织化学方法,并结合真彩扫描仪计算机图象分析技术,观察内膜增生、白细胞阳性指数及类皮质激素的作用。结果:(1)对照组与服药组各对应时间内移植血管内膜相对厚度无显著性差异;(2)CD_(45)阳性细胞指数在两组之间存在显著性差异,术后1W和2W平均阳性细胞指数分别为14.73±0.38％和3.92±0.42％,术后4W和16W各组均无明显的阳性染色细胞。结论:(1)白细胞在血管移植术后1W和2W各占增生内膜的14％和4％左右,可能在诱导移植术后早期内膜增生和促进术后中后期非细胞性增殖方面具有重要意义。(2)本实验条件下的类皮质激素对移植血管内膜增生无明显抑制作用。
To investigate leukocyte infiltration and its quantitative changes relative to intimal hyperplasia in autologous vein grafts at different periods after surgery. Methods: Twenty-four rabbits were divided into two groups (hormone group and control group) in the femoral vein to femoral artery model. The immunohistochemistry for leukocyte common antigen(Dako-CD45), combined with genuine color scanner and computer image analysis system were applied to reveal intimal thickening, positive cell index(PCI) and effect of steroid hormone. Results: (l)The relative intimal thickness showed no statistically significant difference between the two groups at corespending periods. (2) There was nei-ther any statistical significance between the two groups in PCI. The PCI at 1W and 2W after surgery were 14.73%±0.38% and 3.92% ±0.42% respectively. However. No positive stained cells were identified at 4W and 16W after surgery. Conclusions: (l)Steroid hormone showed no obvious inhibitory effect on intimal hyperplasia. (2)Leukocytes infiltration in thickened intima may play a potential role in indu-cing extracellular matrix formation during the early stage of grafting, thus enhancing extracellular matrix accumulation in the late stage of grafting.
