摘要
以石油醚为溶剂索氏提取,薄层层析定性检测,高效液相色谱法(HPLC)定量分析测试甘草种子、根中角鲨烯。辅助气相色谱/质谱进一步验证角鲨烯的存在。结果表明:甘草种子、甘草根中存在角鲨烯,但含量较低。本试验中建立的HPLC测定角鲨烯方法是可行的,确定的色谱条件为:C18柱,流动相V(乙腈)∶V(甲醇)为60∶40、检测波长为210nm、流速为2mL/min、柱温为30℃。可以考虑将角鲨烯做为甘草中一种新的具有高开发利用价值的物质予以深层次开发利用。
In this study,we evaluated whether squalene exists in Glycyrrhiza uralensis Fisch.The oil in the root and seed of G.uralensis was extracted by Soxhlet extractor with petroleum ether,and then squalene was separated and determined by TLC(thin layer chromatography),HPLC(high performance liquid chromatography) and GC/MS(gas chromatography-mass spectrometry) with selected ion monitoring.The results show that the content of squalene in G.uralensis was lower.The HPLC conditions established in this study,which is suitable to the analysis of squalene,are as follows:C18 column,volume ratio of acetonitrile to methanol 60∶ 40,length of detection wave 210 nm,flow rate 2 mL/min,and temperature 30℃.Squalene can be considered as a new,valuable source in G.uralensis which is worth exploiting.
