摘要
目的:探讨蟾酥对耐药大肠埃希菌的体外抗菌效果及其逆转细菌耐药的部分机理。方法:分别检测水提蟾酥和醇提蟾酥对耐药大肠埃希菌的最小抑菌浓度(MIC),采用PCR方法分别检测与低于一个MIC浓度梯度的水提蟾酥和醇提蟾酥作用前、作用1、5、10天的耐药大肠埃希菌耐药基因(TEM、SHV、DHA、CTX-M-1、CTX-M-2、CTX-M-9、OXA-2、OXA-10、oprD2),同时采用RT-PCR方法检测阳性耐药基因mRNA的表达情况。结果:水提蟾酥和醇提蟾酥对耐药大肠埃希菌的最小抑菌浓度分别为1:4,1:8,耐药大肠埃希菌的TEM和CTX-M-9基因在蟾酥作用前后均为阳性,其余耐药基因在蟾酥作用前后均为阴性,蟾酥作用前及作用1天后,耐药大肠埃希菌的TEM和CTX-M-9的mRNA为阳性,而作用5天后,TEM和CTX-M-9的mRNA检测阴性。结论:耐药大肠埃希菌携带多种耐药基因,蟾酥具有逆转耐药大肠埃希菌的耐药性,其逆转机理可能与其抑制耐药基因表达有关。
Objective：To investigate in vitro antibacterial effect of Venenum Bufonis on drug-resistanct Escherichia coli and partial mechanism of its reversal of drug resistance.Methods：The minimal inhibitory concentrations （MIC） of water extract of Venenum Bufonis and alcohol extract of Venenum Bufonis against drug-resistanct Escherichia coli were detected respectively.After water extract of Venenum Bufonis and alcohol extract of Venenum Bufonis at lower gradient concentration than MIC were treated for 1,5 and 10 days,the resistance genes of drug-resistanct Escherichia coli （（TEM、SHV、DHA、CTX-M-1、CTX-M-2、CTX-M-9、OXA-2、OXA-10、oprD2）were detected by PCR.The mRNA expressions of these genes were also invesitigated by RT-PCR.Results：The minimal inhibitory concentrations （MIC） of water extract of Venenum Bufonis and alcohol extract of Venenum Bufonis against drug-resistanct Escherichia coli were 1：4 and 1：8 respectively.The resisitance genes TEM and CTX-M-9 were detected in drug-resistanct Escherichia coli.Others were not found.The treatment of Venenum Bufonis did not induced the changes of resisitance genes in drug-resistanct Escherichia coli.The mRNA expressions of TEM and CTX-M-9 were found in drug-resistanct Escherichia coli.The one-day treatment of Venenum Bufonis did change the mRNA expressions of TEM and CTX-M-9,but the five-day treatment of Venenum Bufonis inhibited mRNA expressions of TEM and CTX-M-9 Conclusion：Drug-resistanct Escherichia coli carry resistance gene TEM and CTX-M-9.Venenum Bufonis can reverse drug resistance of drug-resistanct Escherichia coli.The effect is related to the mRNA expression inhibition of resisitance genes.
