摘要
目的:探讨11-羰基-β-乙酰乳香酸(AKBA)对人类髓系白血病细胞株HL-60细胞凋亡的影响。方法:不同质量浓度和时间的AKBA处理HL-60细胞后,采用琼脂糖凝胶电泳技术检测凋亡带;采用Hochest33342染色观察细胞的形态学变化;Annexin-V-FITC/PI双染法及流式细胞术检测细胞凋亡率。结果:一定质量浓度的AKBA作用于HL-60细胞即可观察到细胞出现凋亡的形态学改变,DNA凝胶电泳出现了凋亡特征性的DNA小分子降解片段带,流式细胞仪检测早期凋亡率最高可达39.49%,总凋亡率可达99.9%,并呈时间和剂量依赖性。结论:AK-BA能诱导HL-60细胞凋亡,其作用呈剂量和时间依赖性。
Aim:To investigated the effect of AKBA on the apoptosis of human acute myeloid leukemia(AML) cell line HL-60. Methods: HL-60 cells were treated by AKBA at various concentrations for different periods of time.The apoptosis DNA ladders was detected by DNA agarose gel electrophoresis.Morphology of HL-60 cells was observed by Hochest33342 staining.Cell apoptosis rate was determined by Annexin-V-FITC/PI double staining and flow cytometry.Results: When HL-60 cells were treated by AKBA at certain concentrations,the apoptosis morphology and the typical apoptosis DNA ladder were observed and the early apoptosis rate detected by flow cytometry was up to 39.49% and the total apoptosis rate was up to 99.9% in a concentration-and time-dependent manner.Conclusion:AKBA can induce apoptosis of HL-60 cells based on concentration-and time-dependent manner.
