摘要
目的通过体外培养体法考察镇静催眠药(安定、咪达唑仑和苯巴比妥)对阿司匹林酯酶活性的影响。方法采用实验室建立的阿司匹林酯酶体外代谢系统,用高效液相色谱直接进样法测定培养体系中阿司匹林(ASA)和水杨酸(SA)的浓度,以SA/(ASA+SA)比值反应阿司匹林酯酶的活性。通过在培养体系中加入或不加安定、咪达唑仑和苯巴比妥时阿司匹林酯酶活性的变化,分析上述镇静催眠药对阿司匹林酯酶活性的影响。结果安定、咪达唑仑、苯巴比妥对阿司匹林酯酶活性的影响分别为-10.71%(P>0.05)、-27.94%(P<0.05)和8.83%(P<0.05),咪达唑仑对阿司匹林酯酶的活性有一定的抑制作用,苯巴比妥对阿司匹林酯酶的活性有一定的诱导作用。结论咪达唑仑、苯巴比妥与阿司匹林合用时应注意药物间的相互作用。
Objective To study the effects of sedative hypnotics on the activity of aspirin esterase in vitro. Methods The metabolic system of aspirin esterase in vitro was established in the laboratory. The separated plasma specimens were treated with the sedative hypnotics,and then the concentrations of aspirin and its metabolite (salicylic acid)in the plasma were determined by RP-HPLC. The ratio of salicylic acid (SA) and aspirin (ASA) represented the activity of aspirin esterase. Results The effects of diazepam,midazolam and phenobarbital on the activity of aspirin esterase were-10. 71% (P〉 0. 05),-27. 94% (P〈 0. 05)and 8. 83% (P〈 0. 05) respectively. The activity of aspirin esterase was inhibited by midazolam,while it was induced by phenobarbital. Conclusion The interaction between sedative hypnotics and aspirin should be paid careful attention to in clinical application.
