摘要
从河北省微生物多样性研究与重点实验室保存的具有抑瘤活性的粘细菌BD105菌株中分离纯化了抑制人宫颈癌细胞的活性物质。应用分析型高效液相色谱对BD105菌株的发酵产物甲醇粗提液进行梯度洗脱,确定混合发酵产物的最适分离条件,再运用制备型高效液相色谱对不同组分进行分离,大量发酵后制备得到组分I(浅黄色粉末状化合物)。应用MTT法对得到的活性组分进行体外抗癌细胞活性检测,发现组分I对人宫颈癌细胞最高抑制活性达91.785%,体内动物实验也显示出高效低毒的生物活性。
An active stbstance of anti-human cervical cancer was separated and purfied in myxobacteria strain BD105 whick stored in key lab of microbial diversity research and application of Hebei province.The fermentation products of this strain was extracted by methanol and gradiently eluted by analytical HPLC in order to confirm the optimum separation conditions of different components.Then these components were separated by preparative HPLC and the active substance of anti-tumor was tested with MTT assay.Under the guidance of biological activity tracking,a compound named Component I was got which was flavescens powder,and its highest inhibitory activity to human cervical cancer cells achieved 91.785%.Experimentation on animals also showed that this compound had a high efficiency with low toxin.
