By using N 2O synchronized HeLa cells as a model, the effect of PKC activation and inhibition on HeLa cell G 1 to S phase progression was investigated. It was found that this process was remarkably influenced by TPA and GF 109203X treatment, which was the activator and inhibitor of PKC respectively. TPA (100 μg/L) could promote G 1/S progression; on the contrary, it was inhibited by GF 109203X (4 mg/L) treatment. Further study discovered this change was related to PKC activity. TPA treatment promoted PKC activity by 90 0%, and GF 109203X inhibited it by 74 1%. It was shown that MAPK signaling pathway also functioned in this process, which itself might be regulated by PKC activity. In conclusion, it was found that these two signaling pathways cooperated to regulate the progression from G 1to S phase.
