摘要
建立高效液相色谱法测紫叶李叶提取液中花青素的成分及含量的方法。提取用80%乙醇为溶剂结合超声的方法,定性、定量分析使用HPLC法。采用岛津VP-ODSC18(4.6mm×150mm)色谱柱;以4%磷酸∶乙腈(V∶V=88∶12,pH值2.0)为流动相,流速0.8mL/min;柱温:30℃;检测波长:520nm。花青素标准品的进样浓度在0.031~3.100mg/mL的范围内呈现良好的线性关系,R2=0.9999,精密度(RSD=1.954%)高,加标回收率平均值为97.29%。高效液相色谱法精密度高,是测定紫叶李叶中花青素含量的准确、简洁、有效的检测方法。
The components and contents of anthocyanidin in Prunus cerasifera leaves were analyzed by HPLC. The anthocyanidin was ultrasonic extracted by 80% ethylalcohol, separated on SHIMADZU VP-ODS C18 column in 4% Phosphateacetonitrile(pH=2.0,V / V=88∶12) at 30℃ and 0.8 mL/min, and detected at 520nm. The results showed that there was a good linear range of 0.031~3.100 mg/mL (R2=0.999 9), the content of anthocyanidin in Prunus cerasifera leaves was 26.234 mg (RSD=1.954%), the average recovery was 97.29%. In a conclusion, HPLC was an actually, simple, and effective method to detect anthocyanidin.
