摘要
目的:通过观察麻杏石甘汤对A型流感病毒感染小鼠免疫器官重量指数和IL-2、TNF-β蛋白表达水平的影响,探讨该方对A型流感病毒感染小鼠的免疫保护作用。方法:采用鼻腔接种法建立A型流感病毒小鼠肺适应株动物感染模型。实验设麻杏石甘汤组、利巴韦林组、抗病毒颗粒组、模型对照组和正常对照组,经灌胃给药或给生理盐水后,检测小鼠体重、肺指数、脾指数、胸腺指数和肺组织中IL-2、TNF-β的蛋白表达水平。结果:麻杏石甘汤明显提高A型流感病毒感染小鼠的体重、脾重指数、胸腺指数与肺组织中IL-2的蛋白表达水平,降低肺指数和肺组织中TNF-β的蛋白表达水平。结论:麻杏石甘汤通过提高小鼠免疫器官重量指数、调节细胞因子的蛋白表达水平而对流感病毒感染小鼠起保护作用。
Objective:By observing Maxingshigan Decoction(MXSGD) on immune organ weight index of mice which were infected by A-type influenza virus and levels of IL-2、TNF-β,to explore immune protection of mice which were infected by A-type influenza virus. Methods: Use nasal inoculation to establish mouse pulmonary infection model animals by A-type influenza virus. Set up MXSGD group,ribavirin group,Anti-virus particle group,model control group and normal control group. After gavaging drug or normal saline,detection of weight in mice,lung index,spleen index,thymus index,the level of TNF-β,IL-2 in lung tissue. Result: MXSGD can obviously improve mice which were infected by A-type influenza virus:inhibited the rate of weight loss,spleen weight index,thymus index and the level of IL-2 in lung tissue by Immunohistochemical method,and reduce lung weight index and the level of TNF-β in lung tissue. Conclusion: MXSGD by enhancing mice's immune function,regulating a variety of inflammation-associated factor secretion levels to protect mice which were infected by A-type influenza virus.
