葡萄糖酸锌喷鼻剂对小鼠流感病毒感染的保护作用 被引量:4

摘要 目的:考察葡萄糖酸锌喷鼻剂对小鼠病毒性肺炎的保护作用。方法:采用流感病毒A1型(H1N1)鼠肺适应株FM1感染小鼠致肺炎模型,肺炎小鼠随机分为基质对照组,葡萄糖酸锌喷鼻剂大、中、小剂量组(6.4,1.6,0.4 mg·kg^(-1)),Zicam组(1.6 mg·kg^(-1)),利巴韦林组(25 mg·kg^(-1)),共给药5 d,每日3次,观察小鼠的死亡数和死亡时间。另外通过对肺炎小鼠的肺指数,肺病变程度以及小鼠肺部病毒滴度的测定来观察葡萄糖酸锌喷鼻剂的体内抗病毒作用。结果:葡萄糖酸锌喷鼻剂6.4 mg·kg^(-1)可降低流感病毒感染小鼠的死亡率,死亡保护率为54%(P<0.01),1.6,6.4 mg·k^(-1)可延长小鼠存活时间,生命延长率分别为94%,114%(P<0.05,P<0.01);葡萄糖酸锌喷鼻剂3个剂量(0.4,1.6,6.4 mg·kg^(-1))均有显著降低小鼠肺指数和抑制肺部病毒滴度的作用(P<0.01),肺指数抑制率分别为25%,32%,33%。结论:葡萄糖酸锌喷鼻剂有明显抑制小鼠流感病毒性肺炎的作用。 AIM: To study the protective effects of zinc gluconate nasal spray (ZGNS) on viral pneumonia in mice. METHODS: Viral pneumonia model established by influenza A1 virus FM1 (H1N1) in mice. The mice were randomized into six groups: matrix control group, ZGNS high dose group (6.4mg·kg^-1), ZGNS middle dose group (1.6mg·kg^-1), ZGNS low dose group (0.4mg·kg^-1), Zicam group (1.6mg·kg^-1) and ribavirin group (25mg·kg^-1). The mice were treated for 5 d, three times per day. Mortality and survival time were observed. Antiviral activities of ZGNS were tested by pulmonary-index, pathologic changes and virus titer of lung in vivo. RESULTS: Compare with matrix control group, mortality (42 %) decreased significantly by ZGNS at the dose of 6.4 mg·kg^-1 (P 〈 0.01), protective rate was 54 %. Survival time increased at the dose of 1.6 mg·kg^-1 (P 〈 0.05) and 6.4 mg·kg^-1(P 〈 0.01), the life prolongation rates were 94 % and 114 % respectively. Pulmonaryindex and virus titer decreased significantly (P 〈 0.01) in three groups of ZGNS, inhibition ratios of pulmonaryindex were 25 %,32 % and 33 % respectively. CONCLUSION: ZGNS has antiviral activity against viral pneumonia in mice.
