摘要
目的:探讨豚鼠膀胱Cajal间质细胞的原代分离、培养及鉴定方法。方法:断颈处死豚鼠,无菌条件下分离出膀胱组织,采用酶消化法分离细胞,将细胞悬液接种于含干细胞因子(SCF)的细胞培养基中培养。用酪氨酸蛋白激酶受体(c-kit)特异性抗体标记细胞,证实细胞类型。结果:培养后的Cajal间质细胞保持其固有特征,可见胞体为梭形且胞体有两个细长突起,该类细胞c-kit抗体荧光染色阳性。结论:用酶消化法可获得豚鼠膀胱Cajal间质细胞,并在体外生长良好。
Objective:To explore the primary isolation,cultivation and identification of interstitial cells of cajal in guinea pig bladder.Methods: The guinea pigs were killed by cervical dislocation,and muscle tissue was isolated from the bladder under asepsis.Then the enzymatic digestion was used to isolate cells from the preparation muscle tissue.The liquid with cell suspensions was inoculated and fostered in the medium with stem cell factor.Interstitial cells of cajal was confirmed by immumofluorescence of c-kit.Results: The interstitial cell of cajal maintained its inherent characteristics by primary cultivation.We could see that the cell had a spindle cell body with two slender bipolars.Fluorescent staining with c-kit antibody comfirmed interstitial cells of cajal was positive.Conclusion: The enzymatic digestion is available to cultivate interstitial cells of cajal in guinea pig bladder and the cells grow well in vitro.
