摘要
目的通过研究草酸对体外培养的Wistar大鼠肾小管上皮细胞造成的脂质过氧化损伤及柠檬酸的保护作用,以探讨尿路结石的形成机制。方法分离、培养正常雄性Wistar大鼠的肾小管上皮细胞,在原代培养第5天时,将细胞分成3组(空白对照组、草酸组及柠檬酸组),分别加入正常培养液、含有草酸(5mM)的培养液及含有草酸(5mM)和柠檬酸(3mg/mL)的培养液,分别于2h后测定各组细胞培养基中丙二醛的及乳酸脱氢酶含量。结果①草酸组和柠檬酸组细胞培养基中乳酸脱氢酶含量均明显升高,与对照组相比差异有统计学意义(P<0·05);②与柠檬酸组相比,草酸组细胞细胞培养基中乳酸脱氢酶含量更高,两者之间比较差异有统计学意义(P<0·05);③草酸组和柠檬酸组细胞培养基中丙二醛的含量均明显增高,与对照组比较差异有统计学意义(P<0·05);④与柠檬酸组相比,草酸组细培养基中丙二醛的含量更高,两者之间比较差异有统计学意义(P<0·05)。结论草酸对体外培养的Wistar大鼠肾小管上皮细胞可以造成脂质过氧化损伤,而外源性柠檬酸能够减轻草酸对体外培养的Wistar大鼠肾小管上皮细胞造成的脂质过氧化损伤作用。
Objective Citric acid is involved in maintaining cellular endogenous defense by increasing reduced nicotinamide adenine dinucleotide phosphate formations(NADPH) .We examined if citric acid can lessen oxalate induced injury of cultured renal tubular epithelial cells in normal Wistar rats. Methods Renal epithelial cells of normal rats were isolated and cultured respectively. At the fifth day of primary culture, we exposed cultured cells to oxalate(5mM) for 2 hours with or without 3mg/ml citric acid in the medium.We quantified malonaldehyde(MDA) and lactate dehydrogenase(LDH) of the medium. Results The presents of oxalate was associated with significant increase in lactate dehydrogenase(LDH) (P〈0.05). In addition, there was significant decrease in MDA when dtric acid was preset in the medium.Conclusion Oxalate can cause lipid peroxidation damage in cultured renal tubular epithelial cells of normal Wistar rats. And citric acid can lessen oxalate induced injury in cultured rat renal epithdial cells.
