摘要
为建立氟甲砜霉素的免疫分析方法,采用混合酸酐法合成氟甲砜霉素人工抗原(FF-HS-BSA)和包被抗原(FF-HS-OVA),并利用紫外吸收光谱及红外光谱扫描方法检测到人工合成抗原偶联成功。利用合成的人工抗原免疫5只6~8周龄BALB/c小鼠,免疫8次后得到氟甲砜霉素抗血清。利用间接非竞争酶联免疫吸附测定法检测抗体效价为1∶6400,可用于进一步实验研究。
In order to establish the immunoassay method of florfenicol (FF), FF was coupled with carrier protein Bovine Serum Albumin(BSA) and Ovalbumin(OVA) to prepare complete antigen FF-HS-BSA and FF-HS-OVA by mixed anhydride (MA) reaction. UV and IR methods were used to identify synthetic antigen which results showed that the reaction was successed. Five BALB/C mice, 6 -8 weaks old,were immunized with FF-HS-BSA, and the polyclonal antibody(pAb) after 8 times immunized was got. The titer of pAb was detected by indirect ELISA, the titer of FF antiserum was 1 : 6 400,it could be used in the next study.
